Che cos’è il sistema del complemento
Il sistema del complemento è una parteessenziale del sistema immunitarioumano che svolge un ruolo cruciale nella difesa contro le infezioni e nel mantenimento dell’omeostasi del corpo. Si tratta di una rete complessa diproteine plasmatiche e cellulariche interagisce in modo coordinato per riconoscere, marcare ed eliminare agenti patogeni, come batteri e virus. Il nome “complemento” deriva dallasua capacità di “completare” l’azione degli anticorpie delle cellule immunitarie coinvolte nella difesa dell’organismo. Il sistema del complemento può essere attivato attraverso due vie principali: la via classica e la via alternativa.
Attivazione del sistema del complemento
Il sistema del complemento può essere attivato attraverso tre principali vie: via classica, via alternativa. via delle lectine.
1.Via classica: questa via viene attivata quando gli anticorpi IgG o IgM si legano a specifici antigeni presenti sulla superficie dei patogeni. Una volta formatosi il complesso antigene-anticorpo, le proteine del complemento C1q, C1r e C1s si uniscono al complesso ed avviano una cascata di reazioni che porta alla formazione della membrana attaccante del complesso (MAC) sul patogeno.
2.Via alternativa: la via alternativa è costitutivamente attiva a bassi livelli nel nostro organismo come meccanismo di sorveglianza delle superfici cellulari per rilevare eventuali anomalie o invasione da parte dei patogeni. In questa via, alcune proteine del complemento riconoscono direttamente i componenti strutturalì dei microorganismí senza bisogno dell’intermediazione degli anticorpi.
3.Via delle lectine: questa via viene innescata dalla presenza di carboidrati specificì su alcuni microrganismì. Le molecole chiamate lectine legano questi carboidrati e iniziano una serie di reazioni che portano all’attivazione del sistema del complemento.
In tutte queste vie l’attivazione iniziale porta alla generazione di enzimi convertasi che clivano proteine precursorie nel sangue quali il componente C3 in frammentì piu piccoli inclusa l’anafilotossina (C3a), un mediatore infiammatorio, e C3b che si lega alla superficie del patogeno o della cellula alterata per segnalare la sua eliminazione.
Il sistema del complemento svolge una serie di funzioni importanti nell’immunità innata e adattativa, inclusa l’opsonizzazione dei patogeni (marcatura per facilitarne la fagocitosì) , l’aumento dell’infiammazione locale, la citotossicità nei confronti dei microrganismí ed il coinvolgimento nelle reazioni immuno-infiammatorie.
Funzioni del sistema del complemento
Il sistema del complemento può svolgere molteplici funzioni:
1.Lisi batterica: alcune proteine del complemento possono formare complessi che distruggono i patogeni invasivi tramite il processo di lisi (distruzione) della membrana batterica.
2.Opsonizzazione: il complemento può rivestire i patogeni con molecole chiamate opsonine, rendendo più facile per le cellule fagocitiche riconoscerli ed eliminarli.
3.Infiammazione: alcune componenti del complemento possono promuovere reazioni infiammatorie localizzate nell’area infettata o danneggiata, attrarre cellule immunitarie sul sito d’infezione e aumentare la permeabilità dei vasi sanguigni.
4.Eliminazione dei complessì antigen-anticorpi: il complemeento puó favorire l’allontanamento rapido degli antigen-anticorpo complessi che si sono formati durante la risposta immunitaria.
Il sistema del complemento è un componente chiave della risposta immunitaria innata ed è coinvolto in una vasta gamma di processi fisiologici e patologici. Un corretto funzionamento di questo sistema è essenziale per mantenere l’omeostasi e proteggere l’organismo da infezioni e malattie.
